Tryptophan Metabolism and Cancer Progression

Abstract: Intracellular tryptophan (Trp) is catabolized to a large repertoire of metabolites via two major pathways: indoleamine and 2, 3-dioxygenases (IDO/TDO) Trp hydroxylase (TPH) pathways. The catabolites possess diverse biological activities carry out various physiological functions. Several such as kynurenine (Kyn) serotonin promote while melatonin 5-methoxytryptophan (5-MTP) suppress cancer growth metastasis. Cancer cell-derived Kyn enhances evasion immunosurveillance by interacting with cell immune membrane aryl hydrocarbon receptors (AHR), respectively. Serotonin exerts its tumor-promoting through type 1 2 receptors. 5-MTP metastasis common mechanisms, i.e., inhibition p300 histone acetyltransferase (HAT) NF-κB activation, suppression cyclooxygenase-2 cytokine transcription. Both block p38 MAPK signaling pathway. Human tissues express increased levels IDO, TDO monooxygenase (KMO) which are correlated reduced patient survival. In summary, metabolism regulates complex mechanisms. provide valuable lead develop new drugs for chemo-prevention adjuvant therapy cancer.
